I am on Cubase 12 and Mac OS 14.4 (MBP), but I “suffer” from this problem from my first steps on Cubase from version 10 on:
I select my Motu 1248 Interface in “Studio” settings. I save all settings individually: Inputs, Outputs, also Control Room, each one under “default”. When I open Cubase Inputs and CR are connected to the 1248. But I always have to select it under Outputs. It’s really weird and sometimes when some time has passed after the last session, I keep forgetting this and have to find out why I hear no sound. Gets a bit frustrating…
I am not a mac user, but may be this helpful.
- Your motu interface is sleeping from the port you connected it.
- You started Cubase first and connected the motu interface second.
- The motu interface is in use by another application.

I remember this being an issue, yes. It is fixed in the latest release update of cubase 12 though. Have you installed it? Also, flushing your preferences might help. Just rename your preferences folder and test it again. Sometimes older settings get carried over from older versions of Cubase, messing with the saved configurations.
Got the latest version and still the same. I have to rename the preference folder then. But how should I rename it?
just put “_old” at the end of the name, cubase will create a new preferences folder from scratch.
