How delete specific unwanted markings

How can I delete these markings and move the note and “C2” leftward to the barline?

Activate Insert Mode (Global Adjustment of Current Bar), select the rests and press Delete.
Don’t forget to switch off Insert Mode when it’s done!

Now the last bit is to add a line to the right of the first note. How do I do that?

I’m not sure what you did, but what I suggested should normally have left a barline after the first note (by creating what’s called a pickup bar):

Actually, I see what you did: you enabled Insert Mode, but not the Global Adjustment of Current Bar submode.

You can either redo the operation with the correct mode^[1], or (from your screenshot) select the first note, then type Shift+M, c,0.25, and press Enter.
