Why isn’t it possible to load a MIDI VST plugin into Layer 1 (Midichords) and then route that Layer 1 to Layer 2, which contains an audio VST plugin (Omnisphere)?
Or is there another way to do this in VST Live?

@kyru Basically a good idea would be to create a designated track to control the Layer. To do so, simply click on “MIDI Input→Tracks→Create Track” in that Layer.
Now load your plug-in in the newly created track. All MIDI data coming from this track will be directed to the Layer’s MIDI input.
I can’t check if this also works with midiChords as the plug-in isn’t available anymore (and seems to be outdated). Using a simple MIDI keyboard and one of Steinberg’s MIDI plug-ins, however, works as described above.

I can load the MIDI plugin in the layer; that works.
In the MIDI track, I can only load Steinberg MIDI plugins; there’s no Chord Maker there.

As far as I understand it: midiChords tries to be both at the same time: a MIDI plugin and a VSTi(nstrument). That’s the reason why you can load midiChords in a Layer, which only accepts VSTis. So, midiChords will behave like an instrument there.
That’s got nothing to do with midiChords MIDI capabilities!
Due to the unavailability of midiChords I tried to do something similar with Arturia’s Pigments. This plug-in can share MIDI data it generates internally and the plug-in will show up as a designated MIDI input source.
Here’s my workflow I’d suggest in your case:

Insert midiChords in a Track/Layer as a VSTi.

Check if midiCords shows up as a MIDI input for other tracks or Layers.
This works perfectly with Arturia’s Pigments. However, here’s the caveat: If midiChords doesn’t announce its MIDI Out as a proper MIDI input device, it won’t work.
