Preload loads plugins internally once, preload Parts essentially does what you do when you activate a Part manually. Preload is faster and works ok in most cases, but some plugins require more thorough action to have them fully preloaded. If “Preload” works for you (as in most cases), you don’t need “Preload Parts”.
