When comping vocals 3 lanes, and using Melodyne for tuning the lanes keep doubling, ie Cubase plays two lanes at once…
This happens in both Cubase 11 and 12 Pro…

Well behind the scenes the comping and Meloydyne tuning are occurring on different files.
Your workflow will be much smoother if you
- Complete all your comping before tuning
- Use Render In Place (or Export) to create a new clean file that captures your comps
- Use Melodyne on this new file to do your tuning
