How do I make the number of measure per system in Flow 2 be four measures, while keeping the number of measures in Flow 1 at three measures?

You would have to do that manually (because Layout Options apply to all flows in a particular layout).
That said, (forgive me) but I have to ask, why do you want to have casting off at all? After all, none of your individual parts follows any casting off…
I agree with @Janus.
However, a simple way to achieve this is to add a page-worth of system breaks, select them all and then copy/paste in the start of each new frame. I think you are asking for trouble though!
One other thing you could do is use a separate layout for the second flow. I hate doing this though.
Hi @StephenBurch, besides the suggestions above, imagining that you will need that “casting-off” for the parts and since those charts are pretty uniform in the note spacing required (inside one flow) another way to obtain this is to insert a Note Spacing Change at the beginning of each flow (in the Violin part layout, for example), and try increasing/decreasing the value until you have the desired “casting-off”:
You can then do Propagate Part Formatting, from setup mode, so that all your parts will have the same look (yes this creates many system breaks (*), but I think is ok for such charts):
Alternatively you can copy the Note Spacing Changes signposts and paste them in each part and each flow (which probably will take more time).
(Sorry, one more variant: define you Casting-Off in layout options for the whole layout, with the mostly needed casting off (3 or 4 bars, or something else), and then correct manually only the flows that use a different casting off. You can do this after you input all your notes, so you can decide which layout option setting gives less work.)
Hi Janus, I’m not sure I understand your question about why I want to have “casing off” at all. Isn’t the “casting off” thing something that determines the number of measures per line? For this next flow (section) of this exercise set each scale and arpeggio will have four measures. The first set of exercises has three measures for each scale exercise, and the next set (flow 2) will have four measures per scale. If there is a better (easier) way to set each line at four measures for flow 2, I would sure love to learn it. Thanks.
